To determine concentration levels of human granulocyte macrophage colony stimulating factor (GM-CSF) in quantitative terms, Human Macrophage colony Sti is used. Primarily, these concentrations are used in cell culture supernatant, plasma, serum, biological fluids.
With a sensitivity of 1.5-2.0 pg/mL, this is strictly used for diagnostic purposes.
Provided reagents:
There are a number of reagents provided, which has to be kept within a temperature range of 2-8°C. With Human Macrophage colony Sti kit further experiments can be done.

· Calibrator diluent 1
· Calibrator diluent 2
· Substrate A
· Substrate B
· Stop solution
Presentation of samples:
· Cell culture supernatant: With help of centrifuge, particulate matter is to be removed.
· Plasma: It is collected by using heparin and EDTA. After collection for a period of 30 minutes, centrifuged for a period of 10 minutes (4° C).
· Serum: Clotting is to be allowed for a period of 10 minutes at (4° C) before actual experimenting procedure begins.
Reagents preparation:
· Each of the available reagents is to be removed from given temperature and has to be kept at room temperature of (20-25°C).
· To dilute the given solution, 60 mL of Wash Buffer solution is added. It is then mixed in a very good manner and finally put at a stable condition.
· Both the substrate solutions are mixed in 1:1 ratio and kept before actual process begins.
Finally with Human Macrophage colony Sti a good experiment is approved and best results are derived!
